The λmax of Sylimarin in ethyl alcohol and PBS was found to be 286 nm and 322nm, respectively. The drug follows linearity in the concentration range 2–20 μg/ml with a correlation coefficient value of 0.998 and 0.999 for in ethyl alcohol and PBS, respectively. The accuracy of the method was checked by recovery. The % recovery was found to be in the range of 99.54– 100.98% in ethyl alcohol and 98.94– 101.4% in PBS, respectively. The precision of the method was studied as an intraday; interday variations, and repeatability. The % RSD value < 2 indicates that the method is precise. LOD and LOQ were evaluated from signal to noise ratio and they were 0.3 µg/ml and 1 µg/ml, respectively.

FTIR Study
FTIR spectra of silymarin, plain PLO gel and drug loaded PLO gel are showed in Figure 1. FTIR spectra of silymarin loaded PLO gel showed that the characteristics peaks of silymarin appear without any change in the spectrum of the drug. These results indicated that there was no interaction could be detected between the drug and the components of the base.
Drug content
Drug content of the prepared formulations was found to be ranged from 98.2 to 101.2% as showed in Table 2. These results indicated that complete distribution of the drug throughout the PLO gel base.

AD skin patient histology was characterized by thickness of the epithelial layer of the skin, presence of vacuoles or holes and increasing in the inflammatory cells in the dermis layer (Figure 7 A) indicating skin lichenification and inflammation of the skin due to itching. On Contrary, the treated skin patient showed decreasing in the thickness of the epidermis and decrease the inflammatory cells infiltrate into the dermis layer (Figure 7 B). These improvements can be explained according to the results obtained from a previous animal study which indicated that silymarin induced anti- oxidative, anti-inflammatory, immune modulating actions and has been shown to support dermal regeneration.29-31 It can be further stated that the effectiveness of the topical formulation of silymarin-based PLO gel which results from enhancement drug molecules penetration despite of skin lichenification. Local occlusive factor, warmth and hydration produced by PLO gel base are among the factors that can increase silymarin skin penetration.
Some patients in the present study received silymarin free base showed mild, moderate and complete improvement from AD manifestation.
