Aseptic technique is the collective method of working and maintaining pure cultures, which requires awareness of where microbes are (4). To determine the unknown’s morphology is the first step in narrowing down possible organisms. A MacConkey and Mannitol Salt Agar tests were conducted on the organism and revealed the following. No growth on the MacConkey plate resulting in a gram positive bacteria and the Mannitol Salt Agar result showed it could be Staphylococcus or Micrococcus. A dichotomous key and observation can be useful tools in identifying microorganism’s taxonomy as a mold, yeast or bacteria. Microbes can be found everywhere in cold, warm, moist or dry environments and molds, yeast and bacteria each have unique characteristics that set them apart from one another and allow them to survive in these environments. Through macro and microscopic observation, different microbes can be observed through the natural liquid and solid phases and be made into slides to be viewed closely under a microscope. Essentially, a random microorganism can be sampled in a dry environment and be identified through observing unique characteristics that would make the microbe be considered either a mold, yeast or bacterium. The properties of selective and differential media suggest that differences between the bacterial species could be used in this lab to identify these six species but their effectiveness is unknown. The efficacy of identifying a completely unidentified bacterium in a cost-sensitive manner using these test also has yet to be determined. We hypothesized that applying a combination of selective and differential media with biochemical tests would provide a presumptive identification of the first group of six unknown bacteria. We also hypothesized the identification an unknown member of the Enterobacteriaceae family referred to as unknown #53 could be definitively identified using the API 20E and other tests.
