Define a reticulocyte
Reticulocytes are young RBCs specifically stained with a supravital stain, such as New Methylene Blue, in order to enhance their remaining nucleolar detail. This detail consists of organelles which have not yet been pushed out of the cell.
Which domestic species does not have reticulocytes in the peripheral blood?
Equine
How is the reticulocyte observed count obtained?
Number of retic counted/1,000 cells observed
How is the reticulocyte corrected count obtained?
(Patient PCV / normal PCV) x Observed retic %
How is a absolute Retic count obtained?
Retics counted/ 1000 x RBC count
What is considered range of absolute count is considered regenerative?
80,000-100,000mm3
Which reticulocytes are counted, aggregate or punctate?
Aggregate
How many fields should be counted during a reticulocyte count?
10; retic count observed per 1000 rbcs
How is a Reticulocyte stain made?
Incubate equal parts of blood and NMB at room temp for 15 mins, then make a push smear, Air dry and evaluate on oil immersion
Why are reticulocyte counts important to assess?
Gives us information on bone marrow's response to anemia
What is the traditional stain for counting reticulocytes?
New Methlyene Blue
What type of reticulocytes have non-clumping reticulum of 1-4 dots?
Punctate
What type of reticulocytes have large clumps of retuculum and often appear bigger and bluer than surrounding RBCs?
Aggregate
Are reticulocytes are efficient at utilizing oxygen?
No
What is another name for a reticulocyte?
An immature RBC
If an animal had an absolute retic of 90,000, would it indicate regenerative or non-regenerative anemia?
Regenerative
If an animal had an absolute retic of 1,000, would it indicate regenerative or non-regenerative anemia?
Non-regenerative
Obtain the corrected reticulocyte count:
Patient: Canine Patient PCV: 37% Normal Canine PCV: 45% Observed retic count: 2% |
(37% / 45%) x 2% = 1.64
Obtain the corrected reticulocyte count:
Patient: Feline Patient PCV: 28% Normal Feline PCV: 37% Observed retic count: 2% |
(28% x 37%) x 2% = 1.51
Obtain observed reticulocyte count:
20 retics observed out of 1000 rbcs |
20/1000= 0.020 or 2.0%
