USE OF MEDIA

Isolation and Identification of Pseudomonas spp

Also determine ability to produce pigments

OTHER NAME OF MEDIA

Pseudomonas agar

Pseudo F agar

King's B medium

Modified Tech agar

MEDIA COMPOSITION
Peptone -20g

MgCl2 - 1.4g

Potassium Chloride-10g

Cetrimide - 0.3g

Glycerol - 10ml

Agar -15g

SELECTIVE AGENT AND IT'S ACTION
The selective agent is CETRIMIDE.

Cetrimide is an antiseptic which is mixture of different quaternary ammonium salts including Cetrimonium bromide .

Cetrimide acts as a quaternary ammonium cationicdetergent causing nitrogen and phosphorous to be released from bacterial cells other than Pseudomonasaeruginosa.

It inhibits all the other microorganisms

ACTION OF OTHER INGREDIENTS

Gelatin- Nitrogen, Vitamins and Carbon source.

MgCl2 and Potassium Chloride - Enhance pigment production .

Agar - solidifying agent.

Glycerol - supplemented as a source of carbon

INCUBATION CONDITION

Incubate the plates at 37 °C for 18 - 24 hr

CULTURAL RESPONSE

Blue(pyocyanin) , or Yellow -green (fluorescein ) pigment which is seen with naked eyes .

some times UV illumination is used to analyse pigment(Florescence) when organism doesn't show pigmented colony.

EXPECTED RESULT

(for pyocyanin pigment)

EXPECTED RESULT
