- you will investigate the effect of antiseptics/antibiotics on bacterial growth using agar plates, and measuring zones of inhibition. - the most effective antiseptics or antibiotics will have the largest effect on reducing bacterial growth (larger zones of inhibition)

microbiology: apparatus list

• one agar plate of bacterial cultures. < once incubated, seal agar plate with tape, to trap bacteria in.


• five antiseptic solutions


• five pairs of sterile forceps < if you use ethanol to sterilise, it must be kept away from naked flames as it will catch fire.


• marker pen


• chromatography paper with discs from hole punching


• beaker of disinfectant < this is important when dealing with bacteria


• eye protection < wear when using vicrkon solution


• protective gloves < wash hands before and after

microbiology: common mistakes

• don't rush, to minimise contamination you must be methodical and careful


• don't seal with tape all the way before incubation! condensation needs to escape, and air needs to get in.


• zones of inhibition may not be perfectly circular, if this is the case measure the diameter twice (at 90° to each other) and calculate a mean.

microbiology: method

1. wash hands/wear gloves


2. on underside of agar plate, use marker to divide it into five equal sections.


3. using sterile forceps, dip one paper disc into each antiseptic solution and allow excess to drain off . (use a separate pair for each)


4. place disc onto respective section on unsealed agar plate containing the bacteria.


5. partially seal with tape, allowing oxygen to enter


6. place upside down & incubate for 3-4 days at 25°.


7. place used equipment in beaker of disinfectant.

microbiology: analysis/results

-remove agar plate from place of incubation and seal around are with extra tape.


-measure diameter of clear zone using a ruler. record measurements in table.


- repeat at 90°. record measurement in table


-calculate mean diameter, record in final column.
