what is a gene |
it is the entire DNA sequence transcriptional unit required for the synthesis of a functional proteins or RNA molecule. It include coding regions(exons) and control regions and sometimes introns. |
what is an interuppted gene |
a gene in which the coding sequence is not continuous due to the presence of introns |
what is the primary RNA transcript |
the orginal unmodofiied RNA product corresponding to a transcription unit |
what is mature transcript |
a modified RNA transcript. modification may include the removal of intron sequences and alterations to the 5' and 3' ends. |
how can introns be indentified |
introns can be detected when genes are compared with their transcription products by either electron microscopy, restriction mapping or sequencing |
what is cDNA |
it is a single stranded DNA complimentary to an RNA, synthesised from it by reverse transcription in vitro |
what is another way u can test/identify introns |
EM of ovalbumin mRNA hybridised to a single strand of DNA that includes the ovalbumin gene complimentary regions between DNA and mRNA form a DNA-RNA hybrid, non-complimentary regions loop out |
what is a overlapping gene |
a gene in which part of a sequence is found within part of the sequence of another gene |
what is alternative splicing |
otherwise identical polypeptides, differing by the presence of certain regions, can be generated by alternative splicing when certain exons are included or excluded. alternative splicing uses the same pre-mRNA to generate mRNAs that have different combinations of exons. |
what are alternative splice sites |
mRNAs have same 5' and 3' exons but internal exons are different |
give examples of alternative splicing |
1. striated muscle mRNA 2. smooth muscle mRNA 3. fibroblast mRNA 3. brain mRNA |
which RNA polymerase transcribes mRNA |
RNA pol 11 |
